Deadliest submarine disaster in U.S. remembered

PORTSMOUTH, N.H. (AP) ? Family and friends who lost loved ones when the USS Thresher sank 50 years ago joined in tossing wreaths into the water Saturday in an emotional service in remembrance of the 129 Navy crew members and civilian technicians who lost their lives in the deadliest submarine disaster in U.S. history.

Hundreds gathered for the memorial service at Portsmouth High School that concluded with a small group tossing three wreaths into the Piscataqua River. During the service, a bell tolled 129 times.

The event, along with the dedication of a flagpole Sunday in Kittery, Maine, aims to call attention to the tragedy 220 miles off Cape Cod, which became the impetus for submarine safety improvements.

Vice Adm. Michael Connor, commander of the Navy's submarine forces, acknowledged Saturday that the safety upgrades came at a steep cost to Thresher families.

"I've talked a lot about the good that comes from the Thresher and the Thresher's loss, but that's probably not a consolation to the families who've lost a father or a son," Connor told a packed high school auditorium.

The USS Thresher, built at Portsmouth Naval Shipyard and based in Connecticut, was out for a routine deep-diving test when it ran into trouble on April 10, 1963.

The Navy believes the failure of a brazed weld allowed sea water to spray onto an electrical panel, causing an emergency shutdown of the sub's nuclear reactor. The ballast system also failed, preventing the sub from surfacing.

Filling with water, Thresher descended deeper and disintegrated under the crushing force of the ocean. Its remnants rest on the ocean floor at a depth of 8,500 feet.

Don Wise Jr., 59, of Plaistow, N.H., who lost his dad, said the Thresher crew members were doing something special, serving on what was a technological marvel, the Navy's fastest and deepest-diving nuclear submarine.

"They were going deeper and faster than anyone. I always considered my dad a hero and an adventurer," Wise said Saturday. "These memorials are how I connect my children and grandchildren with my dad."

Former Thresher crew member Frank DeStefano, 79, of Orange Park, Fla., said he owed his life to a three-day assignment to Washington that took him away from the submarine during the fateful sea trials.

He said he's happy to see that annual memorial events provide an outlet for families and friends to grieve.

"The only good part about these memorials is that we can help those who were really affected, like the families," DeStefano said. "And it's great to see the children that have come along."

Lynne Lawrence, of Alexandria, Va., whose father, Richard DesJardins, was one of the civilian technicians who died, attended the service with two siblings.

In a recent interview, she described her father as a fun-loving, busy engineer, and said she was sad he didn't get to see his children become adults or meet his grandchildren.

"It's a profound loss that affects you forever, but you grow from it and move on," she said. "Because you don't really have any other choice."

After the ceremony, a rifle team fired shots as the wreaths ? one each for Navy personnel and civilian technicians who died, and one for previous Thresher crew members ? were tossed into the river.

Church: Pastor Rick Warren's son commits suicide

LAKE FOREST, Calif. (AP) ? The Southern California church headed by popular evangelical Pastor Rick Warren announced Saturday that Warren's 27-year-old son has committed suicide.

Warren's Saddleback Valley Community Church said in a statement that Matthew Warren had struggled with mental illness and deep depression throughout his life.

"Matthew was an incredibly kind, gentle and compassionate young man whose sweet spirit was encouragement and comfort to many," the statement said.

"Unfortunately, he also suffered from mental illness resulting in deep depression and suicidal thoughts. Despite the best health care available, this was an illness that was never fully controlled and the emotional pain resulted in his decision to take his life."

Warren, the author of the multimillion-selling book "The Purpose Driven Life," said in an email to church staff that he and his wife had enjoyed a fun Friday evening with their son before Matthew Warren returned home to take his life in "a momentary wave of despair."

Church spokeswoman Kristin Cole said he died Friday night.

Over the years, Matthew Warren had been treated by America?s best doctors, had received counseling and medication and been the recipient of numerous prayers from others, his father said.

Still, he struggled over the years.

"I'll never forget how, many years ago, after another approach had failed to give relief, Matthew said 'Dad, I know I'm going to heaven. Why can't I just die and end this pain?'" Warren recalled.

Despite that, he said, his son lived for another decade, during which he often reached out to help others.

"You who watched Matthew grow up knew he was an incredibly kind, gentle, and compassionate man," Warren wrote. "He had a brilliant intellect and a gift for sensing who was most in pain or most uncomfortable in a room. He'd then make a bee-line to that person to engage and encourage them."

The elder Warren founded Saddleback Church in 1980, according to his biography on the church website, and over the years watched it grow to 20,000 members. He and his wife, Kay, began by holding Bible studies for people who weren't regular churchgoers.

Warren is the author of numerous books and was named the top newsmaker of the year for 2009 by the Religion Newswriters Association, gaining attention with his invocation at President Barack Obama's inauguration that year.

In 2008, the church sponsored a presidential forum with Obama and Republican Sen. John McCain. Obama and Republican nominee Mitt Romney had been invited to a similar forum last fall, but Warren canceled it, saying the campaign had become too uncivil.

As Saddleback has grown over the years, it has spread out from its Lake Forest headquarters, 65 miles southeast of Los Angeles, adding several other campuses and ministries around Southern California.

The church says it now offers more than 200 community ministries and support groups for parents, families, children, couples, prisoners, addicts, and people living with HIV, depression and other illnesses.

Shutting down DNA construction: How senescence halts growth of potential cancers

Researchers from The Wistar Institute explain a new molecular mechanism behind the phenomenon of oncogene-induced senescence. By depriving the cell of the ability to make new nucleotides?the building blocks of DNA molecules?cells can suppress cancer development by forcing a damaged cell into a senescent state, where the cell remains alive yet cannot reproduce.

According to the researchers, their findings may offer a new strategy to strengthen the effects of anti-cancer drugs and chemotherapies.

Their results, which appear in the April 25 issue of Cell Reports (available online now), show how a senescent cell may become cancerous if supplied with a new source of nucleotides. Their experiments were performed using nevi?the technical term for moles?and melanoma cells, but the underlying mechanism may apply to all human cells.

"Oncogene-induced senescence is an automatic mechanism that arrests the growth of normal cells when an oncogene or cancer-causing gene is activated to prevent the progression of these cells into cancer," said Rugang Zhang, Ph.D., associate professor in The Wistar Institute Cancer Center's Gene Expression and Regulation program. "We identified how an oncogene can set senescence into motion by suppressing RRM2, an enzyme necessary for producing nucleotides."

Without the nucleotide building blocks of DNA, Zhang says, the cell cannot multiply. When new sources of nucleotides are introduced, however, the cell goes into overdrive, becoming cancerous.

Zhang offers an analogy comparing senescence to a construction site, where workers continue to build as long as they have a steady supply of bricks. When an oncogene is activated, it is like hiring an excess of workers. A normal cell's typical defense mechanism is to cancel orders for new bricks. When all the excess workers quickly run through their supply of bricks, the construction site shuts down. This is essentially senescence. If you suddenly flood the site with new building materials, the workers go into overdrive, building wildly without supervision. In the case of cancer, this causes the cells to put DNA building into overdrive?starting the endless cycles of cell growth and division that are the hallmark of cancer.


Wistar's Katherine Aird discusses the Zhang Lab's findings on the mechanics of oncogene-induced senescence. In the April 25, 2013 issue of the journal Cell Reports, they demonstrate how DNA damage can cause a cell to become senescent, a state where growth is halted. (A mole on your skin is an example of a senescent cell you see every day.) They show how this happens as cells suppress the ability to generate nucleotides (the building blocks of DNA). When you supply the cell with new sources of nucleotides, the cell will go into a frenzy of multiplication -- a hallmark of cancer.

To explore how oncogene-induced senescence works, Zhang and his colleagues compared human moles (essentially, pre-cancerous lesions in a senescent state) to cancerous melanoma cells. They found that stable, senescent cells experienced a dramatic decrease in the available number of nucleotides. This was linked to the suppression of RRM2, a gene whose protein is important for making nucleotides. Further, they linked this RRM2 suppression to mutations in BRAF or NRAS, proteins that control the cycle of growth and replication within cells that have been labeled "oncogenes" because their mutation is linked to melanoma and other cancers. Indeed, they report that melanoma patients with high levels of RRM2 fare worse, overall.

Since cells that lacked nucleotides became senescent, the researchers wondered what would happen if they resupplied the cell with nucleotides?in essence, providing eager workers more bricks. They found that, even in cells with an inactive RRM2 gene, the cell rapidly resumed growing and dividing. Such an event in moles, for example, could be what causes melanoma.

"Moles are probably the most visible example of the effect of oncogene-induced senescence in human cells," said Katherine M. Aird, Ph.D., lead author of the study and postdoctoral fellow in the Zhang laboratory at Wistar. "The cells within a mole may have arrested growth, but they are still alive, and may regain activity, even turning cancerous. That is why your dermatologist might tell you to keep an eye on a seemingly benign mole, as changes in its size, color or shape could indicate that it is no longer benign."

According to Aird, if you could stabilize senescence, perhaps by targeting RRM2, it could put the brakes on even drug-resistant cells.

"This mechanism also suggests a strategy for patient therapy," Aird explained. "If we can decrease RRM2 activity, it could force tumor cells into a stronger senescent state, perhaps improving the effectiveness of chemotherapy or targeted drugs."

Dead star warps light of companion red star, astronomers say

Apr. 5, 2013 ? NASA's Kepler space telescope, in concert with Cornell-led measurements of stars' ultraviolet activity, has observed the effects of a dead star bending the light of its companion red star.

The findings are among the first detections of this effect -- a result predicted by Einstein's theory of general relativity -- in binary, or double, star systems.

The dead star, also called a white dwarf, is the burnt-out core of what used to be a star like our sun. It is locked in an orbiting dance with its partner, a small "red dwarf" star. While the tiny white dwarf is physically smaller than the red dwarf, it is more massive. When the white dwarf passed in front of its star, its gravity caused the starlight to observably bend and brighten.

"This white dwarf is about the size of Earth but the mass of the sun," said Phil Muirhead, Ph.D. '11, of the California Institute of Technology and lead author of the findings to be published April 20 in the Astrophysical Journal, titled "Characterizing the cool KOIs: A mutually eclipsing post-common envelope binary."

"It's so hefty that the red dwarf, though larger in physical size, is circling around the white dwarf," Muirhead continued.

The research team used Cornell-led ultraviolet measurements of the star called (Kepler Object of Interest) KOI-256 taken by the Galaxy Evolution Explorer (GALEX), a NASA space telescope operated by Caltech. The GALEX observations were conducted by Cornell researchers Jamie Lloyd, associate professor of astronomy and of mechanical and aerospace engineering; Kevin Covey, former postdoctoral associate now at Lowell Observatory; and Lucianne Walkowicz of Princeton University and Evgenya Shkolnik of Lowell Observatory.

Still in early phases and for which Cornell students are now being recruited by Lloyd, the GALEX program measures ultraviolet activity in all the stars in the Kepler field of view -- an indicator of potential habitability for planets.

Graduate student and co-author Jim Fuller also did a theoretical analysis of the star system in the context of its future and past evolutions.

The red dwarf orbits the white dwarf in just 1.4 days. This orbital period is so short that the stars must have previously undergone a "common-envelope" phase in which the red dwarf orbited within the outer layers of the star that formed the white dwarf, Fuller explained.

Moreover, the short orbital period means the red dwarf's days are numbered: In a few billion years, the intense gravity of the white dwarf will strip material off the red dwarf, forming a hot accretion disk of in-falling material around the white dwarf.

"This system is especially exciting because it allows us to accurately characterize the peaceful state of these systems before the violent mass-transfer phase begins," Fuller said.

Kepler's primary job is to scan stars in search of orbiting planets. As the planets pass by, they block the starlight by miniscule amounts, which Kepler's sensitive detectors can see.

So far, Kepler has identified more than 2,700 planet candidates. Still ongoing is the mission's search for planets similar to Earth in size and temperature that orbit a star like our sun. Ultimately, Kepler will reveal how common Earth-size planets are in the Milky Way galaxy.

To learn more about this particular star system, Muirhead and colleagues also used the Hale Telescope at Palomar Observatory near San Diego. Using a technique called radial velocity, they discovered that the red dwarf was wobbling around like a spinning top. The wobble was too big to be from the tug of a planet. That's when they knew they were looking at a massive white dwarf passing behind the red dwarf, rather than a gas giant passing in front.

One of the consequences of Einstein's theory of general relativity is that gravity bends light. Astronomers regularly observe this phenomenon, often called gravitational lensing, which has been used to discover new planets and hunt for free-floating planets.

In this new study, scientists used gravitational lensing to determine the mass of the white dwarf. By combining this information with all the data they acquired, they were able to accurately measure the mass of the red dwarf and the physical sizes of both stars.
